Effect of rs17817449 of the FTO gene in the incidence of obesity and type 2 diabetes mellitus in Iraqi women

Abstract : Type 2 diabetes mellitus (T2DM) and obesity were chronic metabolic disease and the main cause of other disease such as heart disease and cancer. Previous studies have proven the role of the FTO gene in the development of obesity. Ninety Iraqi women aged from 25-45 years were enrolled as a samples in this study collected from teaching hospital and the women and children hospital of Al-Diwaniyah province from October to December 2020. Sixty women were suffered from obesity and T2DM and 30 were apparently healthy considered as a control. Tetra- ARMS molecular technique was used to detect the rs17817449 genotypes and allele frequency. Study data to analyzed by Chi-square, Odds Ratio (OR), LSD, P-value and Duncan multiple ranges test used to detect the significance of the means. The results showed significant decreasing in HOMO-IR and Uric acid values in obesity study group compared with control (2.00 ±0.19 versus 4.43 ±2.19; P-value =0.0456) , (4.78 ±0.25 versus 5.37 ±0.18; P-value =0.0044) respectively. While the HBA1c values was increased significantly in obesity + diabetic group compared with control (7.56 ±0.38 versus 4.32 ±0.15; P-value =0.0001). Genotype results of rs17817449 TG heterozygous was associated with diabetes compared with control (66.66 versus 40% ; χ2 = 4.28 ) for TG. The results of this study concluded rs17817449 TG heterozygous was associated with incidence of diabetes in Iraqi patient women.

Padiatric BCG Vaccinee and IgE Responses

Abstract : Fifteen from each of the study groups selected on the bases of simple random sampling were blood collected and sera saved. Total IgE assay was done to each serum in duplicate for each of the individuals forming these test groups using the ELISA IgE Kit. Minimum IgE concentrations for the BCG vaccine groups were found to be lower than that of the nonvaccine control group. This may be attributed to the presence of a tolerogenic epitope ensemble within the BCG antigenic structure. BCG vaccine groups were showing higher IgE concentration means than the control groups. A finding which can be attributed to the presence of atopic allergenic epitope in BCG structure The non-scar bearing BCG vaccinee was showing higher IgE concentration means than those of scar bearing BCG vaccinee. Though these higher mean values may be statistically non-significant. BCG scar fading may be due to the waning of responsible cytokine responses in scar formation as the time post-vaccination prolonged. BCG vaccination in childhood initiates variable degrees of IgE responses among vaccine.

Association of the leptin gene (rs7799039) polymorphism with infertility in Iraqi women

Abstract : Obesity and Type-2 diabetes mellitus regarded chronic metabolic disease and main causes of infertility in women. obesity lead to increase of Leptin hormone level which synthesis by adipose cells and enterocytes and cause systemic hormonal imbalance. Ninety Iraqi women were enrolled as a samples in this study collected from Women's Hospital in Karbala City and teaching hospital and the women and children hospital of Al-Diwaniyah province from October 2020 to January 2021. Sixty infertility women were suffered from obesity and T2DM and 30 were apparently healthy considered as a control. Allele Specific molecular technique was used to detect the LEP rs7799039 genotypes and allele frequency. Study data to analyze by Chi square and odds Ratio and p-value test used to detect significance of the means. genotype result rs7799039 related with GG homozygous associated with obesity compared with diabetes (0 versus 40%; X2 =15, p-value = 0.01) and obesity compare with control (0 versus 26.66%; X2 =9.23, p-value = 0.03). result of rs7799039 related with GA heterozygous associated with obesity compared with diabetes (86.66 versus 40%; X2 =14.06, p-value = 0.0005), and obesity compare with control (73.33 versus 86.66%; X2 =1.66, p-value = 0.20). result of rs7799039 related with AA homozygous associated with obesity compared with control (13.33 versus 0 %; X2 =4.28, p-value = 0.03), and diabetes compare with control (20 versus 0%; X2 =6.66, p-value = 0.01). the result of this study concluded that rs7799039 revealed that GA heterozygous, GG and AA homozygous are associated of incidence infertility that suffered from diabetes and obesity in Iraqi patient women.

CLOSURE OF BASAL CELL CARCINOMA DEFECTS IN THE NASALIS REGION AND THE DEXTRA INFRA ORBITALIS REGION WITH A COMBINATION OF FOREHEAD FLAP, ROTATION FLAP, AND SKIN GRAFT PARAMEDIAN TECHNIQUES: A CASE REPORT

Abstract : Basal cell carcinoma (BCC) is the most common type of skin cancer and frequently appears in the centre of the face, particularly the nose. Long-term exposure to ultraviolet radiation is a risk factor for BCC. Biopsy findings routinely confirm the clinical diagnosis. Complete surgical resection is most commonly used in the treatment of facial skin cancer, and the methods used for defect closure are primary closure, local flaps and skin grafts. This article reports a 73-year-old man who complained of a blackish spot on the side of the nose and infra-orbitalis dextra that occurred about two years ago. The complaint was accompanied by itching and bleeding when scratched. The patient was diagnosed with basal cell carcinoma from the history, physical examination, dermoscopy examination, and histopathology. The surgery is carried out in two stages: first, Modified MOHS surgery and secondly, defect closure is performed using the Paramedian Forehead Flap, Rotation Flap and Skin Graft. On the 28th day of follow-up, the surgical wound had dried up completely. The combination surgical technique of paramedian forehead flap, rotation flap, and skin graft can be a treatment option in large facial basal cell carcinoma to achieve an excellent cosmetic and functional outcome.

Serum PCSK9, lipid profile and fasting blood glucose levels in a samples of Iraqi patient and role in developing of atherosclerotic coronary artery disease.

Abstract : Pro-protein convertase subtilisin kexin-9 (PCSK9) is a protein that regulates the expression of low-density lipoprotein receptors (LDLR) and has critical role in the initiation and progression of the atherosclerotic process. This study aimed to quantify the concentration of PCSK9 protein, total lipid profile and fasting blood glucose (FBG) levels in the serum sample of Iraqi patient with atherosclerotic coronary artery disease (ASCAD) and compared to healthy control groups. The present study include 150 participant (100 patient with ASCAD and 50 control), their ages ranged from 30 to 65 years, the PCSK9 levels estimated by ELISA technique, while lipid profile and FBG determined through routine laboratory methods. The results showed that was a high PCSK9 levels in a patient group comparison to healthy controls group (P= 0.00), while lipid profile serum levels (TC, TG, LDL-C, VLDL and HDL-C) and fasting blood sugar displayed a highly significant difference were compared between patients with ASCAD and control group. According to the findings of serum PCSK9 levels may be consider a better predictors for coronary artery disease.
